چکیده

Influenza viruses are responsible for several pandemics and seasonal epidemics pose a major public health threat. Even after outbreak, the emergence of drug-resistant influenza can disease control problems. Here we report novel 6E3 monoclonal antibody capable recognizing binding to H275Y neuraminidase (NA) mutation, which has been associated with reduced susceptibility NA inhibitors. The had K D 72.74 ?M wild-type 32.76 pM NA, suggesting that it identify pandemic H1N1 (pH1N1) virus. Molecular modeling studies also suggest high-affinity this pH1N1 NA. This was subject dot-blot, enzyme-linked immunosorbent assay, bare-eye detection, lateral flow assay demonstrate its specificity pH1N1. Furthermore, immobilized on Au nanoplate nanoparticles, enabling surface-enhanced Raman scattering (SERS)-based detection mutant Using antibody-mediated SERS immunoassay, virus be detected at low concentration 10 2 plaque-forming units/mL. We in human nasopharyngeal aspirate samples, 6E3-mediated potential diagnostic application. anticipate newly developed SERS-based immunoassay will contribute diagnosis improve treatment strategies patients. • against developed. Evaluation of Factors Influencing Antibody Reduction for Development of Antibody Drug Conjugates

Background: Reduction/alkylation is one of the leading strategies for the development of antibody drug conjugates (ADCs). Precise control of the reduction process would not only yield a defined number of free thiols per antibody but also result in development of more homogenous conjugates. Methods: In the present study, we investigated the effect of various dithiothreitol (DTT) concentrations, ...

Neutralization enzyme immunoassay for influenza virus.

A neutralization enzyme immunoassay (N-EIA) was developed for the detection of antibody titer rises in sera of patients infected with influenza A (H3N2) virus. In this N-EIA, a selected strain of influenza A (H3N2) virus was added to monolayers of LLC-MK2 cells in microtiter plates. After 24 h, the replicated virus could be demonstrated with a virus-specific enzyme-labeled monoclonal antibody. ...

Emergence of drug-resistant influenza virus: population dynamical considerations.

Given the considerable challenges to the rapid development of an effective vaccine against influenza, antiviral agents will play an important role as a first-line defense if a new pandemic occurs. The large-scale use of drugs for chemoprophylaxis and treatment will impose strong selection for the evolution of drug-resistant strains. The ensuing transmission of those strains could substantially ...

APPLICATION AND ADAPTATION OF DOT IMMUNOASSAY TO DETECT AND MEASURE RUBELLA VIRUS ANTIBODY

A dot immunobinding assay ( DIA) was used for a quantitative and qualitative assay of rubella antibody. Purified antigen and conjugated anti-human immunoglobulin (RAHlg) were prepared. Nitrocellulose paper dotted with the antigen was added to serially diluted sera or blood samples. The reacting antibodies were visualized by a peroxidase system. Development of a colored insoluble substrate ...
